While restriction enzyme digestion and ligation of DNA into a plasmid vector has been carried out in molecular biology labs for decades, results can sometimes be difficult to discern. Blue-white screening is commonly used to detect successfully transformed cells, and it is important in any experiment to select the correct colonies. In this GoldBio tutorial, we will show you how to improve the technique, preventing vague results so you can easily select your transformed colonies. While X-Gal will dissolve in other solvents, we strongly recommend using DMF for maximum product stability. It is also beneficial to use a higher concentration of X-Gal. Doing so will yield deeper blue colonies, allow faster incubation and provide easier distinction between successful and unsuccessful colonies.
