In the early morning hours of October 1st, mere hours before the destruction of Raccoon City, which by now became desolate ruin populated mostly by monsters, George Hamilton finds an note by his collegue, Peter Jenkins in one of the few remaining survivors' shelters, asking him to come to Raccoon University. While George heads off alone, Alyssa Ashcroft follows him alongside Yoko Suzuki and RPD officer Kevin Ryman, with three survivors discovering Peter dead as someone had shot him through the head and finding an series of notes revealing that Peter worked alongside another researcher named Greg to create an anti-viral medicine capable of purging T-Virus from the infected humans, with their work having resulted in creation of an compound called Daylight, although problems arose as Peter realized that Greg had ties to Umbrella Corporation, for which he correctly deducted to have manufactured the virus and, increasingly aware that Greg likely has his own agenda, decided to contact George for help shortly before Greg killed him. In the notes, survivors also discover that synthensis of Daylight requires three main components; Base Progenitor Virus, wasp poison and blood of an creature already infected by T-Virus, with the group procuring the venom and Progenitor Virus by venturing into an hidden facility built deep below the university while T-infected blood proves to be a much bigger challenge as massive, humanoid creature is rampaging around the campus and massacres an team of UBCS mercenaries sent to capture or kill it, with the squad member, Nicholai Zinoviev observing the fight from afar and tagging the creature, an Tyrant-class B.O.W. codenamed Thanatos, with an blood extractor, although it gets away and enters the university building where it proceeds to pursue the survivors who are able to subdue it by electrocuting it, allowing them to collect an blood extractor from its back. However, as they start to synthensize the Daylight, power to the machine cuts off, forcing them to venture into the nearby machine storage room where an power reset switch is located, There, group is confronted by Greg, who is revealed to be Thanatos' creator and that he rebelled against Umbrella and assisted in creation of Daylight primarily because they wanted to greenlight Thanatos for mass production against Greg's wishes, as he believed that singular, "perfect" bio-weapon is enough and voices intention to pit Thanatos against the survivors. However, Nicholai, observing the conversation from the ventilation shaft above the room, guns Greg down and retreats, having already left multiple C4 charges around the building, with the survivors proceeding to reset the power to the synthensis machine and create several doses of Daylight before escaping to the front of the building shortly before the charges detonate, destroying it with the explosion attracting the attention of the passing evacuation helicopter whose crew urges survivors to converge on the university campus' back passage in order for the helicopter to land. However, as the group proceeds towards the landing zone, Thanatos returns, forcing the group to fight it and bring it down once more. With the Tyrant down, group reaches the landing zone, only to be faced by Thanatos once more as he, missing an arm and undergoing additional mutations, faces survivors one last time. In the ensuing fight, Alyssa loads one of the Daylight doses into an ampoule shooter and fires it at Thanatos, with the anti-viral compound reacting with the creature and literally tearing it apart, finally allowing three survivors to leave Raccoon City minutes before an thermobarric missile is fired at it, vaporizing the entire city.
