They are unlocked in Arcade mode.

For Billy you must complete 2 Target Actions per round, if your character is defeated, it doesn't carry over, so you must complete the TAs again.

For Saiki, you must complete 5 per round.

Difficulty doesn't matter, so you can play on very easy if you want (I recommend this for Saiki, it's much simpler).

Also, you don't need to actually land the attacks, if your opponent blocks, it counts anyway.
