Telomere function is linked to telomere length via the proteins that interact withdouble-strand telomere repeats (homodimers ofTRF1 and TRF2), proteins that bind to the 3singlestrand G-rich overhang present at the very end ofchromosomes (POT1), and proteins that interactwith these proteins such as RAP1, TPP1, and TIN2as well as the Ku70/80 complex present at thejunction of single- and double-stranded telomericDNA : telomere “closed”. TPP1 and POT1 form acomplex with telomeric DNA via TIN2 and TRF1/2if the length of telomere repeat tract is sufficientlylong. TRF1 and -2 are known to bend telomericDNA and telomerase access is proposed tobe blocked in a repeat length-dependent manner : telomere “open.” The TPP1/POT1 recruits andstimulates enzymatic activity of telomerase preferentially at short telomeres .
