🧠 Morris Water Maze Test | Evaluating Spatial Memory in Mice
In this video, we demonstrate the Morris Water Maze (MWM) test, a gold-standard behavioral experiment used to assess spatial learning and memory in rodents.

🔬 Principle:
The test is based on the rodent’s natural desire to escape from water. A hidden platform is submerged just below the surface of an opaque circular pool, and the mouse must use visual cues around the room to navigate and remember its location. Reduced time to find the platform across trials indicates improved learning and memory.

🧪 Procedure:
Mice were individually placed in the pool from various starting points.

Each trial lasted a maximum of 2 minutes.

Drugs were administered intraperitoneally (i.p.) 30 minutes before the test.

In the first phase, the platform remained in a fixed quadrant.

In the second phase (reversal), the platform was moved to a new location to test cognitive flexibility.

💉 Treatment Groups:
Normal Saline (control)

Rivastigmine – a cholinesterase inhibitor

Diazepam – a benzodiazepine

Plant Extract – investigational memory-enhancing agent

📊 Results – Initial Phase:
Normal Saline: Took over 2 minutes, required manual guidance.

Rivastigmine: Found the platform in just 5 seconds.

Diazepam: Took 2 minutes 31 seconds, also needed guidance.

Plant Extract: Found the platform in 20.7 seconds.

🔄 Reversal Phase (platform location changed):
Normal Saline: Still struggled, 2:11 min, manually guided again.

Plant Extract: Adapted quickly, found the new platform in 7 seconds.

Diazepam: Slight improvement, took 1:21 minutes.

Rivastigmine: Performed well again, found the new platform in 31 seconds.

🧠 Conclusion:
Rivastigmine showed the most consistent and powerful enhancement of memory and learning.

The plant extract demonstrated promising cognitive effects, especially in adapting to the reversal phase.

Diazepam impaired learning, consistent with its sedative properties.

Normal saline-treated mice showed poor memory and required guidance.

This experiment highlights the usefulness of the Morris Water Maze in testing cognitive enhancers and potential treatments for memory-related disorders.
