This particular fight/combo will be available if you sent Jinzaemon Kumano to Doujun in the Abandoned Dungeon when you encounter him the 2nd time before jumping into the Ashina Depths and after you've completed the rest of Doujun's requests following that.
You can fight these 2 in a gank fight where Kumano basically acts like one of the late-game heavy samurai and Doujun basically acts like a Senpou Temple monk. The problem here is the arena. It's small and if you swim back to the Underground Waterway idol I start at in this video, you will crash the game. I think it might be safe to go back to the Ashina Castle - Abandoned Dungeon Entrance idol if you've unlocked the door to Doujun's "clinic", but I didn't test that.
To make things easier on myself, I was trying to find ways to stealth kill one of them, thereby making the fight trivial. The way I found is simply to use either Gachiin's Sugar or Spiritfall to up your stealth and quickly dispatch of Kumano, trivializing the fight. Maybe it's not as manly, but at the end of the day, they are dead.
