There are many techniques for analyzing changes in DNA methylation. The method used depends on factors including but not limited to the availability of the DNA, total number of targets being analyzed and the desired specificity. Bisulfite modification of DNA is the foundation for the majority of assays geared toward analyzing changes in DNA methylation patterns. Bisulfite modification converts non-methylated cytosines to uracils, which are then converted to thymines during DNA amplification by PCR. Methylated cytosines are protected from bisulfite modification. Bisulfite conversion together with sequencing techniques allow a per-nucleotide analysis of DNA methylation patterns.
