Percentage of ionized form of weak electrolyte at particular pH of the solution is important to assess its activity. For example, if a weakly acidic drug is significantly ionized at a particular physiological pH, the drug permeability is reduced across the membrane of the tissue. So for a significant absorption, the drug should be in unionized form but at the same time, for dissolution and solubility, it should be in ionized form. So it is a sensitive balance between ionized and unionized forms that determines bioavailability and distribution of the drugs into tissues.
