An improved freehand sectioning method for small, delicate and dry plant specimens

Описание к видео An improved freehand sectioning method for small, delicate and dry plant specimens

Common histological techniques for observing internal plant tissues involve procedures that change the natural structure of the tissue. Freehand sectioning is an easy method that can produce adequately thin sections for microscopic observations of plant materials in their natural state. However, the commonly used freehand sectioning method has limitations, particularly for dry specimens and when material is limited. An alternative freehand sectioning method has therefore been developed. In this method, the specimen is placed on a flat surface, while the index finger of one hand is placed over it and a razor blade is held in the other hand. The nail of the index finger provides support and control over the razor blade. The slight movement of the index finger determines the cutting spot on the specimen. This allows very fine sectioning of a wide range of sampling materials with minimum tissue loss in a short time.
