New preclinical research suggests that aged garlic extract may help protect the aging brain by improving memory and reducing anxiety-like behavior.
In a long-term study published in Biomedical Reports, University of Missouri researchers fed middle‑aged mice a diet supplemented with aged garlic extract for 40 weeks and then evaluated their behavior and brain biology.
Mice receiving aged garlic extract showed better performance in memory and learning tasks, including novel object recognition and spatial learning in a Barnes maze, while general health, weight, and sensorimotor function remained unchanged.
The supplemented mice also displayed reduced anxiety and neophobia in tests such as the light–dark transition and emergence tasks, spending more time exploring brightly lit, open areas instead of remaining sheltered.
Proteomic analysis of hippocampal and cortical tissue revealed hundreds of differentially expressed proteins in the hippocampus and dozens in the cortex, involving pathways linked to synaptic signaling, apoptosis regulation, cognition, and anxiety-related behaviors.
These molecular shifts included changes in proteins related to tau, amyloid precursor protein, presenilin 1, and brain-derived neurotrophic factor, suggesting potential relevance to neurodegenerative processes such as those seen in Alzheimer’s and Parkinson’s disease.
Researchers propose that the neuroprotective effects of aged garlic extract may stem from its antioxidant, anti-inflammatory, and multi-target actions, which have previously been associated with improved cardiovascular health and reduced oxidative stress.
However, the authors emphasize that the work was conducted in a single male mouse model, without mechanistic validation, and that translation to human therapies remains uncertain and requires further clinical research.
