The NanoBiT system, a bioluminescent structural complementation reporter, uses two subunits of NanoLuc luciferase that are fused to proteins of interest.

Interaction of these proteins brings the subunits together, forming an active enzyme that emits a bright luminescent signal. Initially developed for live cell assays, Dr Ismail has adapted NanoBiT into a biochemical format for primary high-throughput screening (HTS), with subsequent live cell applications for secondary screening.

This video highlights the assay's sensitivity to detect both strong and weak PPIs and its robustness, making it suitable for HTS applications in drug discovery.
