Electrical wave propagation in patterned HEK cells. Electrically HEK cells were grown on patterns of fibronectin to form small islands and serpentine stripes (here, the stripes connect outside the field of view). The cells also expressed a blue light-activated optogenetic actuator (CheRiff). Pulses of blue light evoked action potentials, which then propagated via gap junction-mediated conduction. The voltage is imaged using a far-red voltage-sensitive dye (BeRST1). These experiments showed that the geometry of the sample played a critical role in defining the action potential properties: the spiking in the small islands and the long stripes had very different properties, even though the cells in both cases expressed exactly the same ion channels.
