This movie is part of a PhD conducted by Or Shahar at the lab of Dr. Michal Goldberg (http://bio.huji.ac.il/goldberg/main.htm) in collaboration with Dr. Eran Meshorer (http://meshorerlab.huji.ac.il/).
Shown are Homologous Recombination repair Induced (HRind) cells. A picture was taken every 8 minutes for 2 days (60X, Oil). An Endonuclease (ISceI) fused to the red fluorescent protein, Cherry, and the Glucocorticoid Receptor ligand binding domain (Cherry-ISce-GR) is cytoplasmic. Hence, at the beginning of the movie the nuclei are not red. Following Dexamethasone (Dex) addition, the endonuclease enters the nucleus (shown by red nuclei) and cuts the DNA, forming a double-strand break (DSB) in the DNA. Cells in which the DSB was repaired by homologous recombination (HR) express Green Fluorescent Protein (GFP).
