In spite of significant advances in our knowledge of the molecular mechanisms by which tumor development and progression are governed, and the increasing number of targeted therapies in development, the treatment of the majority of cancers remains problematic. The causes of this situation are essentially related to the high variability of tumors, which increases the difficulty in identifying predictive factors of response to therapeutic agents that could guide clinicians in selecting the appropriate treatment regimen for each patient. Hope comes from the design of preclinical trials based on relevant experimental models representing the genotypic and phenotypic profiles of human cancer pathologies. It has now been demonstrated that patient-derived xenograft (PDX) cancer models clearly represent the human situation. This talk will present how such a cohort of PDX models opens up new avenues for evaluating new targeted therapies or biomarkers in experimental phase II trials.
