The fundamentals of Solid Phase Extraction (SPE) sample preparation, including choosing the correct SPE format and using the four basic SPE steps, are discussed. Choosing the appropriate format for your SPE will be based on the analyte, matrix, volume and concentration of your sample. The five basic formats for SPE products, including the straight barrel or syringe barrel, large reservoir capacity (LRC), Bond Elut Jr., 96-well plate, and pipette tip, are discussed. There are four basic steps to SPE. CLWE stands for Condition, Load, Wash, Elute. Conditioning is usually a two-step process and is used to prepare the sorbent for extraction. The Loading step is used to load your sample. Be sure to understand your matrix and basic characteristics associated with your compound(s) of interest. Washing refers to using a solvent or solution to wash interferences from the SPE material, without removing the compounds of interest. Elution refers to the solvent required to elute the compounds of interest from the SPE material.
