The COVID-19 pandemic is still unfolding; the causative agent is SARS-CoV-2.

By May 31st 2021, there are more than 170 million reported cases, and COVID-19 has claimed more than 3 million lives. It is known that antibody responses play critical roles during the infection. To fully decipher the immunogenicity of SARS-CoV-2 infection, it is essential to assess which protein and which part of the spike protein is highly immunogenic in a systematic way.

To meet this need, the Tao Lab constructed a SARS-CoV-2 protein microarray with 21 proteins and a peptide microarray with 197 peptides that fully covers the spike protein. By applying these two arrays, they analysed the antibody responses of 1,051 COVID-19 patients, and generated a comprehensive antibody responses (IgG and IgM) landscape. Through this landscape, they identified potential biomarkers for predicting the severity and outcome of the patients, for accurate diagnosis.

The Tao Lab revealed that the majority of the IgGs against RBD are conformational. Dominant epitopes on the spike protein were identified, the neutralisation activity of antibodies raised against these epitopes were also tested. This landscape will facilitate our understanding of SARS-CoV-2-specific humoral responses and might be useful for vaccine refinement.
