There are a total of 16 Clockwork Castellans to find and destroy in the Resident Evil 4 Remake. There is one Clockwork Castellan per chapter. The Clockwork Castellans will save between playthroughs, so if you miss a few you will only need to get those few in a new playthrough.

However, there is no way to tell which ones you have missed, only how many out of the 16 you have collected.

As soon as you destroy a Clockwork Castellan it is saved, even if you die straight after it. Destroying all the Clockwork Castellans will reward you with the "Revolution Wind-up" trophy / achievement as well as the Primal Knife for use in RE4, which has unlimited durability.
