Our research focuses on DNA and RNA binding proteins that possess disparate nucleic acid and protein-protein interaction domains that are connected by disordered linkers. The inherent flexibility in the architecture enables formation of a wide array of configurations depending on the context of the DNA/RNA. To investigate how individual domains function to impart functional specificity, our group utilizes fluorescence-enhancement through non-canonical amino acids (FEncAA) to capture the dynamics, on-off rates, diffusion, and remodeling. Studies on Replication Protein A will be presented as a example of this approach.
