This weeks Ascendant Challenge can be found in the Chamber of Starlight Lost Sector in Rheasilvea. It's best to clear out that lost sector first, then use your Tincture of Queensfoil. Once inside, you objective is to make your way across the ascendant plane, being wary of a slew of enemies.

As soon as you enter, there will be 3 Hive Champions. You can take them out to get a Purification Ritual, or press forward. You want to move quickly, but also be mindful of all the enemies here. There is a wide range this time around. Along with the Champions, there will be shadow thralls, taken hobgoblins, phalanx, and even shriekers this time. Do your best to clear out the enemies, and progress across the large map.

Keep following the paths of bridges, and you'll come across more Champions. The further you go, the more aggressive they will become, which lets you know you're on the right track! Once you reach the end of the paths, there will be a named Hive Champion, surrounded by Shriekers. It seems if you stay in the arena, the Shriekers won't attack, but if you try to cheese by jumping to safety they don't approve and will target you. Either way, you should be able to burn down the yellow bar pretty quickly. Supers and heavy work well, as always, but be mindful of any other enemies here like phalanx that can knock you off the edge.
