To mask in After Effects, select a layer, choose a shape tool (Q) or Pen tool (G) from the toolbar, and draw directly in the Composition panel; this creates a mask tied to that layer for isolating or revealing parts of your footage for effects or compositing, with options to adjust path, feather, opacity, and even track it automatically for animation.
