Serotonin is an amine derivative autacoids.
-It is widely distributed amine, present on animals & plants
-In humans, mainly present in GI enterochromaffin cells (90%), platelets and brain.
-Synthesized from tryptophan (in diet) in two steps.
-Platelets do not synthesize but take up from blood (active uptake process in platelets and nerve terminals).
-Cell storage in granules similar to catecholamines (NA and DA)

5HT-1: 5HT-1A, 5HT-1B, & 5HT-1D : GiPCR
5HT-2: 5HT-2A, 5HT-2B, & 5HT-2D : GqPCR
5HT-3 : LGIC (cation channel)
5HT-4: GsPCR
5HT-5: GiPCR
5HT-6: GsPCR
5HT-7: GsPCR
Pharmacotherapy Role:
• Migraine: serotonins produce the migraine due to its vasoconstriction effects; therefore, the methylsergide (5HT-2 antagonist) and sumatriptan (5HT-1 agonist) can effectively control the attack.
• Raynaud’s: release of serotonins from platelet trigger acute vasospastic episodes of larger arteries, ketanserin (5HT-2 antagonist) has prophylactic values in Raynaud’s.
• 5HT-3 antagonist (Ondansetron, Tropisetron, Zacoprid) are widely used in cytotoxic drug or radio therapy induce emesis, as an antiemetic drugs.
• 5HT-4 Agonist/Prokinetics (Cisapride, Mosapride, Renzapride) are used as an antiemetic drugs
