Volume of distribution is a difficult concept because it is challenging to think about how drugs preferentially perfuse from the plasma and into tissues. Drugs will smaller volumes of distribution that match closely the volume of plasma, interstitial fluid, or body water are relatively easy to interpret. Drugs with Vd values in excess of the volume of the human body are more difficult. These high Vd molecules are thought to concentrate in fatty tissue and therefore partition mostly out of the plasma and into other parts of the body.
