A common mistake made during aspiration comes from placing the pipette tip as deep as possible in the source vessel to prevent the aspiration of air. This can lead to excess liquid sticking to the outside of the tip. On top of that, you might aspirate more liquid than intended, because the deeper your tip is submerged during aspiration, the more liquid will enter. Therefore, it's recommended to insert the tip only 2-3 mm into the source liquid. An exception to this is when aspirating extremely low volumes of about 0.2 μl. Here, you should go fairly deep during aspiration to ensure that liquid is actually entering your tip.
