In order to unlock marbles (and challenge #41 Pica Run) you need to use 5 gulp pills in a single run.

I followed the suggestion of a reddit user to start with lazarus and restart the run until you're starting with gulp. Then in greed you can buy all the pills until you get the gulp.

This was my third attempt, in the first two I managed to get only 4/5 gulp 😢.
