Cells communicate by exchanging signaling molecules to instruct each other about their position, function and fate. When a signal is received from a cell, it culminates in the activation of specific combinations of genes, inducing cells to acquire distinct identities and functions.

A relatively small number of signaling cascades is known, and their components have been historically discovered by their epistatic effects. Therefore, it is widely accepted that signaling pathways are conserved linear series of molecular events, irrespective of the cell-type in which they are triggered. However, it is unclear how a small number of conserved signaling cascades can drive the wide-range of differentiation fates occurring during animal development.

The Cantù lab’s experimental efforts are focused on the ‘Wnt signaling pathway’, a molecular cascade important for virtually all aspects of embryonic development, and whose deregulation causes human malformations and several forms of aggressive cancer. To deepen the understanding of this crucial pathway the lab employs sophisticated mouse genetics combined with state of the art high-throughput methods to discover the composition of the protein “arsenal” that allows the activation of the correct genes depending on cell type.

In this presentation Dr. Cantù will provide an overview of how his lab combines CUT&RUN with other technologies to gain new insights into gene regulation during development.

In this webinar we will
provide new insights into gene regulation during development via the Wnt pathway and
discuss the advantages of CUT&RUN to gain insight into this pathway.
