This video provides a tutorial on role of Pyruvate Dehydrogenase Complex (PDH Complex) in Oxidative Decarboxylation of Pyruvate to Acetyl-CoA. PDH complex consists of three enzymes and five coenzymes. E1enzyme - pyruvate dehydrogenase (PDH) bound to thiamine pyrophosphate (TPP), E2 enzyme – dihydrolipoyl transacetylase covalently bound to lipoic acid and coenzyme A and E3 enzyme – dihydrolipoyl dehydrogenase bound to cofactors FAD and NAD+. There are five steps of oxidative decarbodation of pyruvate to acetyl-CoA. The reaction is carried out by PDH complex with E1, E2 and E3 enzymes. Furthermore, PDH enzyme is activated and inhibited by PDH phosphatase (PDP) and PDH kinase (PDK) enzyme, respectively.
