The frequency of daily use of painkillers is dependent on the duration of action of the particular painkiller that you are taking. There are painkiller like paracetamol for which the effect lasts only for 6 hours, so you need to be taking every 6 hours, a pill of paracetamol. Something like ibuprofen, paracetamol combination can work for 8 hours. Do you need to take them for 3 times for 24 hours. There are certain drugs which have duration of action of 12 hours. So you will take tem only for twice a day there are some drugs that are effective only for a single dose and that one single dose is sufficient for 2 hours. So it depends on what is the efficacy and duration of effect for a particular type of painkiller that you are using.
