Since I am the first and only one who tried and did this run, I gonna call it the "exo-run". Since Nioh is a completely different game than Souls-Games in terms of being a real combat/fighting style game, we need surely some different rules. I used the following rules for my run.

What is NOT ALLOWED and makes you lose the run:
Blocking is not allowed (2 exceptions, see further down)
Getting hit by anything what causes damage makes you lose the run!
Being staggered even without getting damage makes you lose the run!
Using living weapon makes you lose the run!
Using a Katana to fight humanoid bosses makes you lose the run!
Playing online makes you lose the run!
Using the Hidden Teahouse before going into the DLCs makes you lose the run!
Getting any environmental damage makes you lose the run!

What IS ALLOWED under certain conditions:
Following attacks can be blocked:
#1 Great Centipede Opening attack
#2 Saika Magoichi Opening attack
Getting damage from poison in "The Silver Mine Writhes" - Mission is fine
Getting fall damage is fine
Dashing through fire while not taking damage is fine (water buff prevents fire damage)
