Reversible vs Irreversible Inhibition:
In reversible inhibition, the binding is non-covalent, reversible binding. Here removal of the inhibitor restores the enzyme activity. So the duration of action depends on how fast the drug is removed from the body. On the other hand, in irreversible inhibition, the bond is a strong covalent bond, which permanently inactivates the enzyme. Here the restoration of activity depends on the synthesis of new enzymes, which usually takes longer. So generally, the action of irreversible inhibitors lasts longer. In short, reversible inhibition is like trapping an enzyme for a while, whereas reversible inhibition is like killing it.
