Standard Plate Count Water Test
The standard plate count method is a microbiological test that estimates the number of bacteria in water by growing colonies on a nutrient plate and counting them. This method is used to assess the quality of drinking water and to check the efficiency of water treatment.
The standard plate count (SPC) for drinking water, as described in Standard Methods (American Public Health Association, 1975), is the plating of small quantities (usually 1.0 or 0.1 ml) of a properly collected water sample in a nutrient agar medium (plate count tryptone glucose extract agar) and incubating aerobically .
The standard plate count is where a sample is diluted in a series of dilution blanks, and then aliquots (measured amounts) of the dilutions are plated using either the pour plate method or spread plate method. The expected result is a single bacterial cell gives rise to a single bacterial colony.
