Video S2. In Toto Live Imaging of Mouse Development from Gastrulation to Early Organogenesis, Related to Figures 2 and 7

(A) Two-day light-sheet imaging of mouse development from E6.5 to E8.5 with a ubiquitous nuclear reporter (H2B-eGFP, CAGTAG1 reporter line). The raw multi-view image data produced by the microscope visualized in Video S1A were combined by content-based image fusion and are shown here as maximum-intensity projections for three different views, distal view (top, YZ-projection), anterior-posterior view (bottom left, XY-projection), and lateral view (bottom right, XZ-projection). A visualization of the image data in the form image slices across the volume is provided in Video S2B and S2C. Time is represented in the format hh:mm:ss. (B) Image slices of a nuclear labeled mouse embryo at different developmental stages: Maximum-intensity projections of the anterior-posterior view of an mKate2-nls expressing mouse embryo developing from a late streak to early somite stage (left) and the corresponding series of image slices acquired across the image volume at early, middle, and late time points (t50, t150, and t350) showing the level of cellular resolution that can be achieved in different tissues and germ layers. To minimize file size and reduce playback time, only every second image plane is shown for the middle time point and only every third image plane is shown for the late time point. Time is represented in the format hh:mm:ss. (C) Improving spatial resolution in streak-stage embryos with four-view imaging: Rotating maximum-intensity projections (left) and XY/XZ slices (middle and right) of the image volume of an mKate2-nls expressing embryo, comparing two-view imaging (top, images combined with content-based fusion) with orthogonal four-view imaging (bottom, images combined with multi-view deconvolution). The XY/XZ slices of the image volume demonstrate the improvement in resolution and isotropy achieved with four-view imaging. The video sequentially shows two specific time points of the time-lapse recording, starting with the E6.5 early-streak stage and 11 hr later.
