The Periodic Acid–Schiff (PAS) stain is a classical and widely used histochemical technique for the demonstration of carbohydrates and carbohydrate-rich tissue components. The principle of PAS staining is based on the oxidation of vicinal glycol groups in carbohydrates by periodic acid to form aldehydes, which subsequently react with Schiff reagent to produce a characteristic magenta coloration.
PAS stain highlights neutral mucins, glycogen, basement membranes, fungal cell walls, and glycoproteins, while acidic mucins remain negative unless combined with Alcian Blue. It is of great diagnostic importance in conditions such as glycogen storage diseases, diabetes mellitus (renal changes), mucin-secreting tumors, glomerular basement membrane pathology, and fungal infections.
Due to its simplicity, reliability, and broad diagnostic application, PAS staining is a high-yield and frequently examined topic in histopathology for B.Sc. MLT students in Nepal.
