ABTS [TEAC] - Trolox-Equivalent Antioxidant Capacity Assay - Principle, Advantages & Limitations

Miller and Rice-Evans were the first to report the ABTS•+ assay in 1993, The assay is also known as TEAC [Trolox-Equivalent Antioxidant Capacity].

Initial Method:
The generation of a radical cation (ABTS•+) was done by reacting metmyoglobin (Met-Myb) with hydrogen peroxide (H2O2) to produce hydroxyl radical (HO•).
Improved Method:
ABTS•+ radical generated by treating with Ammonium or Potassium persulfate in the ratio 1:0.5.
Oxidation of ABTS reaction is long process and requires 12-16hrs of incubation.

Principle:
The TEAC Test (Trolox-Equivalent Antioxidant Capacity) is based on the reaction with the colored and relatively persistent 2,2 ′-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS•+ ) radical cation which has a strong absorption band at 734 nm.
