Thank you for joining us today for this introduction to Visium HD 3'. Both Visium HD and the brand new HD 3' offer subcellular resolution. HD is probe-based, and the off-the-shelf kits are available for human and mouse. HD 3' is poly-a capture, opening up your ability to use it for all eukaryotes. In any spatial biology experiment, tissue quality is absolutely critical for success.

For all spatial biology, we require a DV200, a RIN, and an H&E so we can assess the tissue's morphology.

For Visium HD, we recommend FFPE samples due to their extensive stability. Visium HD 3' has only been validated for fresh frozen by 10x Genomics. I'm sure they are working on other sample types.

There are two sample areas per every slide, and you can use a standard microscope slide.

For sequencing, Visium HD and HD 3' are validated on the Illumina NovaSeq X+. 10x recommends 275 million paired-end reads per sample area. Note that this will likely only sequence the library to 40% depth, but you can always sequence more deeply after you've reviewed your data.

10x Genomics offers two free softwares, Space Ranger and Loupe Browser, you can download from their website. The output from Space Ranger can be imported into Loupe Browser, and the image you see on the right is Loupe Browser. And that concludes our introduction to Visium HD 3'.
