Platypus Reclayed retains the same core gameplay from its original source material, where the player-controlled aircraft from Mungola sets course towards the heart of Colossatropolis. As with any other game in the series, the main goal is to destroy hordes of enemies while avoiding getting hit by enemy fire. The scoring system is fundamentally identical to the original version, but whenever the player's score is lost due to using continues, the previous score will be displayed at the bottom of the current score to keep track of the player's high score, until the new score exceeds the old score. Furthermore, there is also an indicator bar for the amount of points required for gaining extra lives.

The player is given the option to select one of several different ship types to use before starting a run. Up to two players can play the game through local co-op, similar to the original, but the game can be opted with both players complete with any chosen compatible device before starting the game. Additionally, the second player joining in mid-game is still possible, as with the original version, but will be unable to select their craft, as it will automatically choose the previously used ship type from a previous session. Unlike in the original game, however, no credit is used through either options, and will be spent only whenever a player rejoins.
