However, due to their heterogeneity in size and composition, EVs are difficult to isolate and classify. In this roundtable, Stefan Wild (Senior Scientist R&D, Miltenyi Biotec) moderates the discussion about research progress and challenges on the clinical application of extracellular vesicles (EVs). Three leading experts in the field, Tony De Fougerolles (CEO, Evox Therapeutics), Joana Correia (CEO, Exogenus Therapeutics) and Joshua Welsh (Research Fellow, NIH) share their experience on the technical limitations of EV detection, EV engineering methodologies, advantages of EVs in comparison to cellular therapy, and the imperative need for standardized EV analytics.

Due to their physical and biochemical properties, EVs are ideal biological vehicles, as they show low immunogenicity and the ability to cross biological barriers. Moreover, the potential for targeted delivery to specific tissues or cells combined with the advancements in EV engineering methodologies, opens endless possibilities for EV applications in disease treatment. While the interest in EV research has been progressively increasing, technology that allows for precise and reproducible detection and characterization of EVs will be essential for the implementation of their promising clinical applications.

A reliable and robust method allowing analysis of multiple markers is required to study the surface signature of EVs and identify potential subtypes with clinical relevance. To provide an easy and fast screening of EV surface proteins, we developed the MACSPlex EV Kit IO, a multiplex bead-based flow cytometry assay. The combination of 39 different MACSPlex EV Capture Beads allows for the detection and analysis of 37 markers plus 2 isotype controls in parallel, by standard flow cytometry. MACSPlex EV Kit IO provides you with the possibility to analyze established EV detection markers in combination with EV epitopes that are specific for the immuno-oncology (IO) field of research.
