Splitting the signals between GFP and YFP on a traditional flow cytometer usually requires a special filter set. Here, we have data collected from a 4 lasers/15 parameters Fortessa and we wanted to see if the unmixing algorithm adds anything to the method already suggested by Marcus and Raulet (https://doi.org/10.1002/cyto.a.22347). Short answer is no. But the video doubles as a tutorial on using the unmixing algorithm in FlowJo, so it's all good.
