So we are back with another senescence video this week because i just love learning about it (sort of my job :D) and have been recently given that it was my lab meeting this week. Anyway, in this video we take a look at a recent study that explores how differing oxygen partial pressures (basically how much O2) influences the senescence associated secretory phenotype (SASP). So to question could hypoxia reduce the SASP?

Why? Well, oxygen is really important, I think we can all agree on that, however, you may not know that different tissues in your body are exposed to very different partial pressures of oxygen. Cells however are clever and have ways of sensing changes in oxygen. Key to this response is a protein known as HIF-1 (hypoxia inducible factor 1). Activated by hypoxic conditions HIF-1 enables cells to adapt to low oxygen. As a consequence of these adaptations, energy levels drop in a cell and this activates AMPK, a protein you may have heard of since it is activated by metformin...Anyway, AMPK is known to repress mTOR signalling. mTOR has been shown to influence the SASP. So, yeah, this is where things get interesting.

But why? Well, understanding how oxygen influences our body is quite important given that there is interest in using oxygen as a therapeutic strategy whether mimicking low oxygen levels with drugs or exposure to high oxygen partial pressures such as with hyperbaric oxygen therapy. So is high or low oxygen good..? Well, haha, yeah, its confusing and it seems either short exposure or finding the "sweet spot" is going to be best..thing is, don't think we are quite there yet.
