I actually have no clue what relationship those pups have to Gattuso. It could just be the leader of the pack, the mother, or anything really. It just comes and goes. It's known for being a tough battle in the early parts of the game. It is arguably still a tough battle in New Game Plus on Unknown Mode depending on your level and set-up. I quite like Gattuso.

Also, here's the tweedles for a second round. Unlike the first encounter at the castle, this doesn't count as a boss battle. I'm pretty sure losing here counts as a game over unlike the first encounter too. I don't know why, but it is what it is, I guess.
