Dr. Mindbender developed Serpentor as the leader of the terrorist group COBRA. In order to build this being, COBRA combed the world for traces of Vlad Tepes, Rasputin, Napoleon Bonaparte, Julius Caesar, Hannibal, Genghis Khan, Attila the Hun, and Alexander the Great. He was never meant to be Cobra Commander's replacement. He was supposed to be a more iconic figure to motivate the troops. The intention was for Cobra Commander to retain his power.
