Gist: Once CAR-T derived from difference preprocessing steps, these CAR-Ts were co-culture for stimulation and checking the cytokines release by each of them. Before that, which CAR construct is optimum is also validated and find out the third generation 28BBz iCAR-TCTL gives more cytokine production.
Studies were conducted in presence of antigen and in the absence of antigen and find out that the immature CAR-T gives cytokines production in the absence and presence of antigen whereas mature CAR-T and pCART (CAR from PMBC) produce cytokine in the presence of antigen. Thereafter, they did the in-vivo studies with each CAR engineered with luciferase that will show the signals once the CAR-T are activated. Results show that CAR from the immature T cells didn’t give any signals (neither at tumor specific site nor at control) whereas the CAR-T from the mature T cell lineage and PBMC show luciferase signal at the antigen site with the intensity of the signal is highest in the later. So far these results indicate that T-cells from PBMC perform better than T cells derived from iPSC.
