Here's a general procedure for preparing Sabouraud Dextrose Agar:
Ingredients:
Sabouraud Dextrose Agar Base
Dextrose (glucose)
Agar-agar
Distilled water

Procedure:
Weighing Ingredients:
Weigh the appropriate amount of Sabouraud Dextrose Agar Base according to the manufacturer's instructions.
Mixing Agar Base:
In a large container, mix the Sabouraud Dextrose Agar Base with distilled water. Stir to dissolve the agar base completely.
Autoclaving:
Autoclave the mixture to sterilize it. Follow the autoclaving parameters recommended by the manufacturer. Typically, this involves heating the medium to 121°C for 15 minutes.
Cooling:
Allow the autoclaved medium to cool to approximately 45-50°C. Be cautious not to let it solidify during cooling.
Adding Dextrose:
Aseptically add the appropriate amount of Dextrose (glucose) to the medium. Stir well to ensure even distribution.
Pouring Agar Plates:
Pour the agar mixture into sterile Petri dishes to solidify and form agar plates.
Solidification:
Allow the agar plates to solidify at room temperature.
Storage:
Once the agar plates have solidified, store them in the refrigerator until ready for use. Make sure to label them appropriately with the date and contents.
