Pyrazinamide is a medication used to treat tuberculosis. For active tuberculosis, it is often used with rifampicin, isoniazid, and either streptomycin or ethambutol. It is not generally recommended for the treatment of latent tuberculosis. It is taken by mouth.
Formula: C5H5N3O
Molar mass: 123.113 g/mol
CAS ID: 98-96-4
Boiling point: 357 °C
Trade name: Rifater, Tebrazid, others
Metabolism: liver
Pyrazinamide inhibits renal excretion of urates, frequently resulting in hyperuricemia which is usually asymptomatic.
Pyrazinamide is an antibiotic used to treat tuberculosis (TB). Pyrazinamide is available in generic form. Common side effects of pyrazinamide include: nausea
Moa
Pyrazinoic acid binds weakly to aspartate decarboxylase (PanD), triggering its degradation. This is an unusual mechanism of action in that pyrazinamide does not directly block the action of its target, but indirectly triggers its destruction.
Common side effects of pyrazinamide include:
nausea,
upset stomach,
vomiting,
loss of appetite,
mild muscle or joint pain, or.
fatigue.
Resistance
Mycobacteria are known to acquire resistance to the antituberculous drug pyrazinamide (PZA) through mutations in the gene encoding pyrazinamidase (PZase), an enzyme that converts PZA into pyrazinoic acid, the presumed active form of PZA against bacteria.
There are 6 disease interactions with pyrazinamide which include:
gout.
hepatotoxicity.
liver disease.
renal dysfunction.
diabetes mellitus.
hemodialysis.
