"पुलिस और मुज़रिम फिल्म की कहानी डीएसपी विशाल खन्ना (विनोद खन्ना) को कॉलेज के बाहर दो गुड़ों की पिटाई से शुरु होती है। ये गुड़ें गृह मंत्री धर्मपाल सिंह
(सदाशिव अमरापुरकर) के होते हैं, जो मंत्री के रुप में जनता की सेवा करने के बजाय अपने काले कारनामें करता है। बॉम्बे के पुलिस कमिश्नर वीर बहादुर सिंह (राज कुमार) अपनी पत्नी शारदा और बेटी ज्योति के साथ रहते हैं। जब डीएसपी विशाल खन्ना दो कुख्यात लुटेरों और दलालों, शेरा (शेर खान) और जग्गा (महावीर शाह) को गिरफ्तार करता है और जेल में बंद कर देता है, तो कुछ गुंडों के बॉस बनारसी दास ज्योति का अपहरण कर लेता है, उसे फिरौती के लिए पकड़ लेता है और अपने गुंडों की रिहाई के बाद ही उसे छोड़ता है। विशाल उसे बचाने का बीड़ा उठाता है और काफी हद तक सफल भी होता है, जिसके बाद शेरा और जग्गा को दोषी ठहराया जाता है और दो साल की जेल की सजा सुनाई जाती है। उनकी सजा पूरी होने के बाद, शेरा और जागा बनारसी दास (चरन राज) से मिलते हैं और वीर बहादुर सिंह और विशाल खन्ना की ज़िंदगी को उलट-पुलट करने का एक रप्लान बनाते हैं।
