In spite of significant advances in our knowledge of the molecular mechanisms which govern tumor development and progression, most novel therapies still fail upon reaching phase III clinical trials. This occurs even though millions of dollars are spent on target validation and drug optimization in preclinical models. There is an obvious need for better humanized and more clinically predictive models of human cancer. The combination of large collections of highly characterized patient-derived xenograft (PDX) in vivo models mimicking cancer heterogeneity and 2D or 3D ex vivo primary culture assays should now help the development of more effective targeted therapies and are now available for clinicians in personalizing the treatment of their patients. In this webinar, we will discuss how the association of ex vivo assays, in vivo PDX model collections and refined preclinical pharmacology improve clinical predictability, as well as the challenges for increasing their relevance for specific cancer pathologies.
