This is how I write up to 7 ISO formatted sample CDs to a single SD card for use with SCSI2SD. In this example I show the process with an Akai S3000XL but I believe this method should work for E-mu, Roland, and other samplers (provided you use a sample CD which matches the proprietary sampler format, ie an E-mu sampler reading an E-mu format ISO).
