CLIA- Chemiluminescence Immuno Assay
This test is used for detection of Antibody.
Abbreviation: Antibody - Ab
Antigen - Ag
Principle: This techniques is similar to ELISA. The reaction carried out in two steps.
STEP I:- Primary Ab present in the sample binds with Ag.
Suppose a person is HIV positive and we have to detect the presence of Ab.
The HIV caused by gp41 Ag, So the infected person produces gp41 Ab.
Ag pre-coated coated well is taken in which gp41-Ag is present. As the blood sample of infected person is poured in the well the Ab gp41Ab binds with gp41Ag and form a Ab-Ag complex.
STEP II :- HRP Horseraddish Peroxidse labelled secondary Ab which is antihuman gp41 antibody binds with Ab-Ag complex.
Reagent containing LUMINOL derivatives along with Electron Transport Agent binds to HRP labelled secondary Antibody.
As it binds HRP-Ab oxidised the Luminol and emits Photons.
Photons are the packets or units of light energy.
The released Light energy is detected by Luminometer.
Intensity of Light is directly proportional to the concentration of Antibody present in the blood sample.
More the binding of HRP-Ab with Luminol more the oxidation more the production of light more is the concentration of Antibody in the blood sample.

Pre-coated well containing gp41 Ag binds with Primary Antibody Present in blood sample binds with HRPlinked Secondary Antibody now it binds with LUMINOL derivatives with electron transport agent results in Releases PHOTONS.
