In this video, we walk you through the detailed process of preparing a semen sample for Intrauterine Insemination (IUI). Proper preparation is crucial for improving sperm quality and maximizing the chances of a successful IUI procedure.

Here’s a step-by-step breakdown of the IUI preparation process:

✅ Media Preparation:
We use two types of media: the lower and upper media. Both are warmed to 37°C to ensure optimal conditions.
The lower media is added to the test tube first, followed by the upper media. The upper media is carefully slid down the test tube wall to prevent it from mixing with the lower media.

✅ Semen Sample Preparation:
Before beginning, we check the semen sample by placing a drop on a slide and viewing it under a microscope. This helps assess the quality and motility of the sperm.
The semen sample is then added to the test tube by sliding it down the side, again ensuring it doesn't mix with the media.

✅ Centrifugation Process:
The sample is placed in the centrifuge for 10 minutes at 1,500 RPM to separate the sperm from debris and other unwanted particles.
After centrifugation, the supernatant (the top fluid) containing debris is discarded. What remains is a concentrated sperm pellet at the bottom of the test tube.

✅ Washing the Sperm:
The sperm pellet is mixed with washing media and centrifuged again for 3 minutes at 650 RPM. This additional washing process ensures the sperm is clean and ready for use.
Once complete, the supernatant is discarded, leaving the bottom layer with the best sperm for IUI.

✅ Final Step: Loading the IUI Catheter:
After the sperm is processed, we check the sperm count to ensure optimal quality.
Finally, the prepared sperm is loaded into the IUI catheter, ready for insemination.
