Clean The Microphone and Speaker holes on the iPad
If you find that one of your iPad’s microphones sounds muffled after you test it or it has no sound at all, let’s clean them. My favorite way to clean iPad microphones is by using a dry, un-used toothbrush to clean the microphone grill at the bottom of your iPad and the small black dot microphone to the right of the rear-facing camera. Simply slide the toothbrush over the microphones to dislodge any stuck pocket lint, dirt, and dust.

To turn up the volume of an iPad microphone, you should: Click Start→Control Panel→Sounds and Audio Devices. Open the Control Panel→click Sounds, Voices and Audio Devices→click on Sounds and Audio Devices.

To enable microphone on iOS (iPad, iPhone, iPod) go to Settings → Privacy → Microphone on your device. Find Yousician on the list and enable microphone access by tapping on the slider (so that it turns green). If the microphone is not working, check instructions from Apple for troubleshooting.

Also have a look at this step, it is not part of the video:
Get Rid of Any Screen Protectors and Cases
Chances are the new screen protector or the case you have recently installed on your device is interfering with the microphone, and thus making it not work on your device. Remove any of the third-party cases or covers you may have put on your device, as they could be the culprit of the issue. Once the cases are all removed, see if you can use the microphone normally.

Launch the Settings app on your iPhone and navigate to Accessibility - Audio/Visual. Switch off Noise Cancellation toggle. Have a call with a friend and make sure the microphone is functioning properly. If it's still malfunctioning then re-enable the feature and move to the next trick.
