“Bottom-Up” proteomics has become a standard tool for molecular biology, separating peptides via liquid chromatography en route to a tandem mass spectrometer that measures the masses of particular peptide ions and then revealing their sequences through dissociation followed by measurement of fragment ion masses. Measuring intact proteins through “Top-Down” methods in tandem mass spectrometry, however, has been much less widely adopted. This talk will explore the advantages of intact “proteoform” measurement for biology and examine some of the challenges that have been a barrier to its adoption.
